Challenges of Accurate Sub-Microliter Pipetting
As sample volumes fall below one microliter, effects that are negligible in larger transfers can become dominant. Reliable miniaturization depends on both the metering mechanism and the complete laboratory workflow.

Why fractions of a microliter are difficult
An absolute error of 0.05 µL is a modest proportion of a 10 µL transfer but a large share of a 0.2 µL target. Mechanical play, seal behavior, meniscus motion, tip wetting and operator timing all become more visible when the intended sample is tiny.
Liquid properties change the result
Viscosity, surface tension and volatility influence how a liquid enters and exits a tip. A method calibrated for aqueous buffer may not behave identically with a viscous reagent or solvent-rich mixture. Evaporation, temperature and dwell time can also distort a small-volume test.
Aspiration and delivery are separate engineering problems
A system may aspirate a tiny amount reproducibly yet leave part of it hanging at the tip. The result is an accurate intake but an inaccurate transfer. DRD’s differential design aims to combine a finely resolved aspiration stroke with an independent, higher-flow delivery action.
Designing a meaningful validation
Test across several nominal volumes and plate positions, document the liquid and consumables, and use an appropriately sensitive measurement method. Record bias, coefficient of variation, failed deliveries and carryover. Demonstration results should not automatically be treated as specifications for every application.
For a system-level decision, compare these principles against the performance data and technical documentation supplied for the particular DRD product. See the related DRD product and discuss the intended application with the engineering team.

Frequently asked questions
What is sub-microliter pipetting?
It refers to transferring volumes below 1 µL, such as 0.5 or 0.2 µL.
Why can a hanging drop matter?
At very small volumes, residual liquid on the tip can represent a substantial fraction of the intended dose.
Are published performance numbers universal?
No. Liquid, volume, tips, hardware, calibration and test conditions must accompany any performance claim.
